چکیده

Background: Non-specific immunotherapy using Bacillus Calmette-Guerin (BCG) is currently the preferred treatment to prevent non-muscle-invasive bladder cancer (NMIBC) recurrence after surgery. However, it remains suboptimal as recurrences and progressions are respectively observed in 60% and 20% of cases. Among the most promising alternative approaches to BCG is the blockade of immune checkpoints. Using the MBT-2 murine bladder cancer model, we analyzed the phenotype of the tumorinfiltrating immune cells, and their expression of various immune checkpoints such as PD-1, CTLA-4, LAG-3, and TIM-3. We hypothesized that blocking PD-1/PD-L1 pathway would boost the anti-tumor response.

Antitumor effects of lipoxygenase inhibitors on murine bladder cancer cell line (MBT-2).

Arachidonic acid (AA) metabolites derived from both the lipoxygenase (LOX) and cyclooxygenase (COX) pathways transduce a variety of signals related to cell growth. Selective blockade of the different metabolic pathways of AA (using a general LOX inhibitor NDGA, a 5-LOX inhibitor AA861, a 12-LOX inhibitor baicalein and a general COX inhibitor ibuprofen) revealed that murine bladder cancer cell l...

Recombinant interleukin-2 enhanced the antitumor effect of ADV/RSV-HSV-tk/ACV therapy in a murine bladder cancer model.

BACKGROUND Previous studies demonstrated the antitumor effects of IL-2 and ADV/RSV-HSV-tk in bladder tumor models. In our study, we employed the intramuscular injection of recombinant IL-2 combined with ADV/RSV-HSV-tk gene therapy in the MBT-2 murine bladder tumor model. MATERIALS AND METHODS In the in vitro study, after adenoviral gene transduction efficiency had been assessed, the cytotoxic...

Tumor Establishment Features of Orthotopic Murine Bladder Cancer Models

PURPOSE Animal tumor models are important for the evaluation of novel therapeutic modalities. Since the initial report of an orthotopic bladder tumor model, several modifications have been proposed to improve the tumor take rate. Here we compared the HCl-pretreated and electrocauterization-pretreated orthotopic murine bladder tumor models. MATERIALS AND METHODS MBT-2 murine bladder cancer cel...

Ganglioside G(M3) overexpression induces apoptosis and reduces malignant potential in murine bladder cancer.

We demonstrated previously (S. Kawamura et al., Int. J. Cancer, 94: 343-347, 2001) that large amounts of ganglioside G(M3) accumulate in superficial bladder tumor, compared with invasive bladder tumors and that exogenous G(M3) inhibits the invasive potential of bladder tumor cells. To apply the G(M3) overexpression system to bladder tumor therapy, direct evidence for the important role of G(M3)...

Anti-tumor effects of anti-PD-L1 therapy in an orthotopic bladder tumor model

Antibodies that block PD-1/PD-L1 interactions have shown efficacy against both lung and skin cancers in early-stage clinical trials, and may also be effective in other tumor types, particularly bladder tumors. PD-L1 expression has been previously reported to correlate with high-grade tumors, a high recurrence rate, and reduced survival rate in patients with bladder cancer. These findings and th...
